A Comparative Study on the Ovarian Functions and Embryo Developmental Capacity in Immature Rats Treated with Two Different Gonadotropins(PMSG and Purified FSH)

Abstract
Immature rate of an age of 28 days received a superovulatory dose(40IU) of PMSG(SOV-PMSG); a physiological dose(4IU) of PMSG(control) or a continuous a c. infusion with a mixture preparation of purified 60IU FSH/25IU hCG(SOV-FSH). Some of females were sacrificed at 72hr after gonadotropin treatments to assess the yield and oocyte quality together with ovarian steroid responses. The rest of other females were caged with fertile males at 80hr after the gonadotropin treatments, scored for the occurrence of mating on the following morning and subgroups sacrificed on days 2 through b of pregnancy to evaluate the yield, gives normality and developmental capacity of preimplantation embryos coupled with circulating steroid profiles. Although mean counts of recovered oocytes were not significantly different between the two groups of SOV-PMSG(59.1¡¾2.3 oocytee/female) and SOV-FSH(81.1¡¾1.7 oocytes/female), infusion of purified FSH/hCG caused a significant(p$lt;0.01) increase in the mean percentage of normal oocytes(95.8%) ovulated at 72hr after gonadotropin treatments A majority of superovulated oocytae following this treatment regimen exhibited a nuclear maturation stage synchronized to metaphase it (90.0%). In contrast, superovulated oocytee obtained from the rats of SOV-PMSG group displayed premature or markedly asynchronized anclear maturation stages at prophase I (10.7%), anaphase I (22.0%), telophase I (19.9%) and metsphase I/II (35.1%). Ovarian contents of 17¥â-estradiol and androgens, elevated greatly above control values, in the rats of SOV-PMSG group were significantly(p$lt;0.01) reduced following SOV-FSH treatment regimen. Compared to SOV-PMSG group, infusion of purified FSH/hCG greatly prevented both the early loss of preimplantaion embryos and the developmental retardation which took place after day 2 of pregnancy onwards following the SOV-PMSG treatment regimen. It was also associated with a significant(p$lt;0.01) increase in the mean percentage of normal embryos observed on each day of pregnancy examined: 94.7% vs 62.4% on day 2, 79.0% vs 30.2% on day 3, 78.1% vs 11.8% on day 4 and 81.8% vs 7.2% on day 5. The circulating levels of 17¥â-estradiol, androgens and progesterone in the SOV-PMSG group was significantly(p$lt;0.01) elevated above control values from days 2 to 6 of pregnancy. In contrast, partiularly, the elevations of circulating 17¥â-estaadiol and androgen levels in the SOV-FSH group were significantly(p$lt;0.01) reduced on days 2 to 4 of pregnancy, compared to SOV-PMSG treatment regimen.
The overall results of the current study suggest s novel method of infusion of the purified FSH/hCG preparations as a new superovulation agent, establishing its usefulness for investigations that often require large numbers of viable good quality oocytes or embryos. This study also illustrates the close association between the high yields of normal oocytes and developing embryos and the low levels of circulating 17¥â-estradiol and androgens during the pre- and poet-ovulatory periods in the rat.
